  • CONTEXTE GENERAL Recherche et compréhention des mécanismes de la dérégulation de la fonction des cellules immunitaires, pour y remédier par des thérapies immunomodulatrices, dans des conditions pathologiques. L'immunothérapie est de plus en plus souvent envisagée, ou déjà utilisée, pour ces pathologies. Il s'agit d'un domaine en pleine expansion et il est important que des Equipes algériennes se positionnent sur ce sujet, en partenariat avec des Equipes internationales. Les développements technologiques attendus peuvent être considérables.
  • TRAVAUX RECENTS Nos travaux actuels sont focalisés plus particulièrement sur les cellules de l'immunité innée (cf. Immunomodulation of innate immune cells response and cell-based immunotherapy).

Modèle 2 : monocytes en co-culture avec les cellules autologues primaires humaines du cancer du sein (ER-/PR-/HER2+)
Modèle 3 : monocytes classiques (moM1, CD14++CD16-), non-classiques (moM3, CD14+CD16++), et intermédiaires (moM2, CD14++CD16+), isolés de patients lors d’un infarctus de myocarde

Points essentiels [Highlights]  | our New findings

Nous avons montré pour la première fois que :
1. Durant l’infection à S. aureus, le traitement à la vitamine C (AscH2) favorise la survie, la fonction et la réparation/l'activation du cycle cellulaire des cellules endothéliales humaines, et inverse la libération d'ICAM-1 soluble et la voie métabolique de l'énergie cellulaire en rapport avec l’absorption du glucose. Aussi, la forme réduite de la vitamine C prévient l’exacerbation de la réponse inflammatoire, tout en induisant l’augmentation de l’activité bactéricide. (PubMed: PMID 33676147URL). 

2. Les biomarqueurs des activités fonctionnelles phénotypiques des monocytes sont modifiés après co-culture avec des cellules de cancer du sein humain primaires (ER-/PR-/HER2+). Le traitement avec la metformine entraîne une libération accrue de la cytokine anti-tumorale IFN-γ et du calcium libre intracellulaire (ifCa2+), et une nécrose cellulaire accrue lors de l'interaction entre les cellules du cancer du sein et les monocytes autologues. 3. L'aspirine immunomodule les activités fonctionnelles des monocytes, favorise leur commutation (switch) vers un phénotype classique (moM1, CD14++CD16-), induit une baisse de l'activité iNOS et une hausse de l'activité de l'arginase lors d'un infarctus du myocarde. (PubMed:  PMID 32172203. URL). 

4. La stagnation cellulaire serait fortement impliquée dans l'apoptose des cellules NK périphériques, ainsi que dans la commutation vers un phénotype régulateur NK Foxp3+ lors de leur interaction avec les globules rouges. Le mouvement cellulaire peut être l'une des approches potentielles requis pour stimuler les activités et la survie de ces cellules au cours d'un cancer solide, en régulant négativement les niveaux d'apoptose associée à p53 et d'expression d'ERK. 5. Le rituximab (RTX, un anticorps monoclonal chimérique anti-CD20) inverse les anomalies des activités fonctionnelles des monocytes, ainsi que leur production de cytokines pro-inflammatoires au début du diabète de type 1 humain. (PubMed: PMID 31322069. URL).

6. La metformine combinée au sélénite de sodium immunomodule le burst respiratoire et la phagocytose du macrophage dans les conditions normales, régule sa capacité de lyse intracellulaire et d'expression de la NADH déshydrogénase (complexe I), et induit sa commutation vers le phénotype "M2"(Arginase, ARG1). La metformine et le sélénite de sodium modulent différemment l'expression de CD14, CD80, CD86 et HLA-DR. 7.  La thymoquinone (TQ; 2-isopropyl-5-méthyl -1,4-benzoquinone, C10H12O2), peut être proposée comme un adjuvant puissant dans une approche vaccinale contre la leucémie aiguë lymphoblastique T, promouvant les réponses des macrophages M1, classiquement activés, pulsés avec des lysats de lignées de cellules Jurkat nécrotiques (NecrJCL), et la production de cytokines antitumorales au cours de l'interaction avec les cellules T CD4+ autologues.
